Opsynvi® (macitentan and tadalafil)
for Pulmonary Arterial Hypertension
Opsynvi (macitentan and tadalafil) is a once-daily oral tablet used to treat pulmonary arterial hypertension (PAH) in adults with WHO functional class II-III. It combines macitentan, an endothelin receptor antagonist (ERA), and tadalafil, a PDE5 inhibitor. Macitentan prevents endothelin-1 from binding to receptors, reducing arterial wall thickening, while tadalafil increases cGMP to relax blood vessels, improving exercise ability and reducing the risk of worsening symptoms and hospitalization.
